Yes, I have the msmtp package in my system packages, and use it from
Mu4e in Emacs.

Sorry, I don’t know. I use a ~/.msmtprc file, since that’s what I had
working before I switched to NixOS.
GitHub - nix-community/home-manager: Manage a user environment using Nix [maintainer=@rycee] has an msmtp module which I use. It works fine. There might be some bug if you define multiple accounts, not sure I got that fixed upstream yet.

I use it with notmuch, but outside configuration.nix. Since it
normally contains users-specific settings (and may include
passwords etc) I imaging it can be handled better via homeManager.
Personally I prefer org-mode, mostly because it’s a super-nice
way to document, describe and share pretty anything on any kind
of system, including things that are not “IT-related”…
